Design Notes

The TEV recognition sequence encoded in the rough center of this part is flanked on both sides by sequences that code for three glycines. This feature was designed to serve as a homologous region allowing for facile substitution of either a receptor or a transactivator around the TEV recognition sequence using the Gibson ligation assembly method.

This part includes human arginine vasopressin receptor 2 (AVPR2), the tobacco etch virus (TEV) protease recognition sequence, the DNA-binding domain from the Gal4 transactivator of Saccharomyces cerevisiae, and the VP16 transactivation domain from the herpes simplex virus.
